Using rush is just like a normal turn without enemies attacking. So the sunbeam will work, as is poison, and the 3-turn bonus effects.

Are you sure? I'm certain there have been instances where the Rush command wasn't available at the beginning of the battle, but the next turn I could use it ...

Yeh, every move you make, besides defending I believe, makes the bar grow. It is possible to use it more than once in a battle.

Actually, you can do Rush only after going through 6 fights. You can beat the fights or run if you want, but either way the bar fills up one sixth for each fight.
One way it's handy is to do some serious leveling up in new areas.
When you get somewhere and the monsters are a little too difficult, spend a while running around running from any fight you can't use Rush in. After a few fights having been won with Rush you should be fine to stand toe to toe with the local baddies.
